Diffuse large B-cell lymphoma (DLBCL) is a malignant tumor. While standard therapies improve cure rates, they remain ineffective for some individuals. Finding novel therapeutic approaches is crucial. Macrophage polarization plays a significant role in tumor development, and recombinant hirudin (rH) has been actively researched for its effects on macrophages.
Qiang Pei and the team aimed to validate rH role in macrophage polarization and PAR-1’s mechanism using clinical samples and a cellular model to support new therapeutic approaches.
The study assessed the expression of macrophage polarization markers, cytokines, and PAR-1 in clinical samples. A cell model was established through co-culture with THP-1 and OCI-Ly10 cells.
Flow cytometry, ELISA, and RT-qPCR were used to evaluate cell polarization and cytokine expression, confirming the model’s success. Different doses of rH were then tested to determine its effect on cell polarization. The role of PAR-1 in macrophage polarization was investigated by transfecting si-PAR-1 and pcDNA3.1-PAR-1.
Researchers found higher expression of M2 macrophage markers (CD163 + CMAF+) and PAR-1 in 32 DLBCL samples. Inducing monocyte differentiation into M0 macrophages and co-culturing with OCI-Ly10 lymphoma cells revealed similar trends in the cell model compared to clinical samples. rH was shown to promote M1 macrophage polarization while inhibiting M2 macrophage polarization. Additionally, PAR-1 was found to regulate macrophage polarization and inhibit cell proliferation, migration, invasion, and angiogenesis.
The study concluded that rH inhibits macrophage polarization toward the M2 type, while PAR-1 regulates macrophage polarization, proliferation, migration, invasion, and angiogenesis in DLBCL.
